TOPIC
▸ Innovative Mapping Technologies for Pre-clinical Studies

❝ In basic and translational research on cardiac and neurological disorders, high-resolution and high-throughput electrophysiological mapping technologies play a critical role. This lecture will introduce the application of innovative optoelectrophysiological platforms in pre-clinical studies of the heart and nervous system, highlighting their functional imaging capabilities across different levels, from cells and tissues to whole organs.
❝ Traditional electrophysiological techniques such as patch clamp and microelectrode arrays (MEA) provide high spatiotemporal resolution but face challenges including technical complexity, limited throughput, and single-signal output. In contrast, dual-channel optoelectrophysiological systems integrate optical sensing of membrane potential and calcium ions, enabling simultaneous recording of action potentials (AP) and calcium transients (CaT) in cardiomyocytes or neurons. Coupled with highly sensitive imaging systems, this approach greatly enhances data acquisition efficiency and multiparametric integration.
❝ Applications of this technology include induced pluripotent stem cell (iPSC)-derived cardiomyocytes, cardiac organoids, tissue slices, retinal sensory layers, gastrointestinal slices, and in vivo open-chest animal models. These studies enable investigation of arrhythmia mechanisms, drug effects on cardiac rhythm and calcium dynamics, and high-resolution cardiac assessments in small animals such as zebrafish. Moreover, experiments such as vagus nerve stimulation and posterior cardiac optical mapping in live animals demonstrate the potential for comprehensive cardiac functional monitoring through integrated electrical and optical approaches..
